Recombinase Polymerase Amplification for Rapid Detection of Zoonotic Pathogens: An Overview

نویسندگان

چکیده

With the advent of molecular technology, several isothermal techniques for rapid detection zoonotic pathogens have been developed. Among them, recombinase polymerase amplification (RPA) is becoming an important technology rapid, sensitive, and economical pathogens. RPA has advantage being able to be implemented in field settings, because method requires minimal sample preparation performed at a constant low temperature (37–42°C). rapidly promising tool detection, prevention, control diseases. This article discusses principles its derivatives, including coupled with lateral flow testing (RPA-LF), real-time fluorescence RPA, electrochemical flocculation their applications

Recombinase polymerase amplification assay for rapid detection of Francisella tularensis.

Several real-time PCR approaches to develop field detection for Francisella tularensis, the infectious agent causing tularemia, have been explored. We report the development of a novel qualitative real-time isothermal recombinase polymerase amplification (RPA) assay for use on a small ESEQuant Tube Scanner device. The analytical sensitivity and specificity were tested using a plasmid standard a...

Rapid Detection of Mycobacterium tuberculosis by Recombinase Polymerase Amplification

Improved access to effective tests for diagnosing tuberculosis (TB) has been designated a public health priority by the World Health Organisation. In high burden TB countries nucleic acid based TB tests have been restricted to centralised laboratories and specialised research settings. Requirements such as a constant electrical supply, air conditioning and skilled, computer literate operators p...

Recombinase Polymerase Amplification Assay for Rapid Diagnostics of Dengue Infection

BACKGROUND Over 2.5 billion people are exposed to the risk of contracting dengue fever (DF). Early diagnosis of DF helps to diminish its burden on public health. Real-time reverse transcription polymerase amplification assays (RT-PCR) are the standard method for molecular detection of the dengue virus (DENV). Real-time RT-PCR analysis is not suitable for on-site screening since mobile devices a...

Development of a Recombinase Polymerase Amplification Assay for Rapid Detection of the Mycobacterium avium subsp. paratuberculosis

BACKGROUND The detection of Mycobacterium avium subsp. paratuberculosis (MAP) infections in ruminants is crucial to control spread among animals and to humans. Cultivation of MAP is seen as the gold standard for detection, although it is very time consuming and labour intensive. In addition, several PCR assays have been developed to detect MAP in around 90 minutes, but these assays required hig...

Development of Isothermal Recombinase Polymerase Amplification Assay for Rapid Detection of Porcine Circovirus Type 2

Porcine circovirus virus type II (PCV2) is the etiology of postweaning multisystemic wasting syndrome (PMWS), porcine dermatitis, nephropathy syndrome (PDNS), and necrotizing pneumonia. Rapid diagnosis tool for detection of PCV2 plays an important role in the disease control and eradication program. Recombinase polymerase amplification (RPA) assays using a real-time fluorescent detection (PCV2 ...
